At the beginning, tetrazolium substance, which is the primary and most important substance of this test, is used in powder form along with PBS substance. By combining these two substances, a solution is obtained that is highly sensitive to light and has a yellow color. It is absorbed by the cell membrane with its positive charge. After these steps, this yellow solution enters the cells with the help of endocytosis and goes to the mitochondria.

This substance can be activated in the mitochondria by succinate dehydrogenase in the respiratory cells of the cell, where it turns into purple deposits. In fact, this problem means that if the cells in the device are alive, it is shown and recognizable because of the succinate dehydrogenase enzyme.

To detect the color of the resulting solution, we use a device called a spectrophotometer. This device is able to determine the optical density of living cells at a wavelength of 579 nm with a base wavelength of 630 nm.

To perform the MTT test, regardless of the main device, we need some equipment. An autoclave is needed to sterilize glass containers. A bain-marie is needed to heat the culture medium and open the frozen sample. We use centrifuges to separate the cells. And we also use an inverted optical microscope to see the cells. Also, in the laboratory environment, we use a class II laboratory hood to prevent the spread of odors or toxic gases.

Important points of the MTT test

  1. It is better to test 3 times for each cell to reduce the percentage of error.
